As in the goo you squirt into the air valve? No, don't use it. Either use a plug kit to plug the tire from the outside, or take it to a shop and have them patch it from the inside. If it's punctured on the shoulder, it can't be patched.

It works by forming a thick gel type substance within the tire, which seals the puncture and inflates the tire. It is important to drive the vehicle after installin the sealant, otherwise it forms a large pile on one side of the tire, causing a severe in balance in the wheel, which may lead to an accident.

Well when you spray the sealant inside the tire it begins to expand. As it does this it pushes against the inside of the tire acting as the air would. This allows you to drive a short distance to repair the tire. Then it turns back to liquid.
