Although there are more meanings, the two most common ones I know are: Biblical: After Cain killed Abel, God cursed Cain. To ensure Cain would not be killed, God marked him to warn others. Vampiric: At some point, the Mark of Cain became a way of saying one was vampire

God cursed Cain for killing Abel; Cain feared that he'd be killed because of that curse, so God "marked" Cain with the promise that Cain's death would be avenged sevenfold. That "look" that some people have that says, "Don't mess with me" is the Mark of Cain. It's not a good thing, today's macho-man ideals notwithstanding. See Genesis 4.
